【摘要】 目的 研究杜仲的不同加工方法对其质量的影响。方法 通过杜仲粗皮占杜仲干燥树皮的质量分数及去粗皮前后浸出率的比较,粗皮与杜仲中松脂醇二葡萄糖苷含量、农药残留量及重金属含量的比较,新鲜树皮发汗前后松脂醇二葡萄糖苷含量的比较等进行研究。结果 粗皮占杜仲干燥树皮质量分数为12.7%(n=3);去粗皮前后的醇溶性浸出率没有显著性差异,水溶性浸出率比未去粗皮高11.26%(P<0.05,n=5);粗皮残留有害物质较高而含松脂醇二葡萄糖苷较少(0.050 4%,n=3);发汗前后松脂醇二葡萄糖苷含量:未发汗生品>未沸水浸发汗样品>沸水浸发汗样品,三者之间均有显著性差异(P<0.05,n=5)。结论 杜仲在原产地加工中“去粗皮”是必要的。值得注意的是:“发汗”后松脂醇二葡萄糖苷的含量却下降。

Abstract:Objective To study on the quality of Eucommia ulmodies processing in planting by different methods. Methods The quantity percent of coarsed cortex, water and ethanol extract were compred, the contents of Pinoresinol diglucoside (PDG) were determined by HPLC, the residues of pesticides and the content of heavy metal element were carried out to investigate the quality. Results The quantity percent of coarsed cortex was 12.7% (n=3). Ethanol extract had no remarkable difference whether scrape the coarsed cortex or not. Water extract was higher 11.26% (P<0.05, n=5) than no scraped. The content of heavy metal element of coarsed cortex was high, but PDG was low (0.050 4%, n=3). The content of PDG of Eucommia Cortex which Fahan had no soaked in boiling water higher than soaked, but lower than no Fahan. There were remarkable differences among them (P<0.05, n=3). Conclusion Scrape the coarsed cortex is necessary of Eucommia ulmodies processing in planting. It deserve to take notice that the content of PDG declined after Fahan.

杜仲是杜仲科植物杜仲Eucommia ulmoides Oliv.的干燥树皮,具有补肝肾、强筋骨、安胎、降血压之功效,产于湖北、四川、贵州、云南等省。杜仲传统的产地加工中,“去粗皮”及“发汗”处理是必不可少的一个环节,本实验对不同的产地加工方法对其质量的影响进行了较为系统、全面的研究。
1 杜仲药材加工质量评价体系的建立
在未知杜仲明确的有效成分前,选择水溶性、醇溶性浸出物来评价其质量。随着研究的不断深入,已明确杜仲中松脂醇二葡萄糖苷(Pinoresinol diglucoside,PDG)有明确的降压作用,绿原酸(Chlorogenic acid,CA)、桃叶珊瑚苷(Aucubin, AUC)等也有一定的降压作用[1]。通过《中华人民共和国药典》[2]2005版(一部)及杜仲相关文献的比较分析,笔者选择PDG的含量及醇溶性、水溶性浸出物作为评价指标,对其进行研究。
1.1 松脂醇二葡萄糖苷含量测定方法的研究
1.1.1 实验材料
1.1.1.1 药材
杜仲药材由上海华宇药业有限公司提供(产地:四川都江堰,批号G2003100803),经南京中医药大学中药鉴定学教研室吴德康教授鉴定为杜仲科植物杜仲Eucommia ulmoides Oliv.的干燥树皮。置鼓风干燥箱中60 ℃干燥1 h,粉碎,密封,备用。
