大孔树脂柱色谱法分离纯化茶皂素的方法研究

摘要::通过吸附、解吸试验,筛选适合分离纯化茶皂素的大孔吸附树脂并确立纯化工艺参数。结果表明,HPD—100型大孔吸附树脂对茶皂素分离效果良好,较佳静态态吸附pH=3.5,在规格为600mm×40mm分离柱中,装样柱高405mm、洗脱液乙醇浓度为80%、洗脱流速为10 mL/min条件下,洗脱效果最好,在此条件下所得茶皂素回收率为71.70%,纯度达到97%左右。

Macroporous resin column chromatography separation and
purification method of tea saponin
Abstract: : The adsorption, desorption tests, screening for isolation and purification of tea saponin and macroreticular resin purification process parameters established. The results show that, HPD-100 macroporous resin type for separation of the effect of tea saponin good, better static state adsorption pH = 3.5, in the specifications for 600mm × 40mm column, the column-like high-mounted 405mm, ethanol concentration of eluate 80%, elution flow rate of 10 mL / min under the conditions of elution best conditions in the tea saponin recovery from 71.70% for the purity of around 97%

茶皂素是一种天然表面活性剂[1],存在于在茶叶、茶籽、和油茶籽中,尤以油茶籽中含量最大。纯茶皂素是白色微细柱状结晶体,分子式为C57H90O26,分子量1203,熔点223-224℃,pH=6.2。分子结构式如下[2]:
